Guardians Of The Galaxy Films
Let's address this option first and foremost, as it will be the option most fans will expect as the game is being developed.
With the movie characters being as popular as they are, there's a good chance the game could follow the outline of the two MCU films.
Chris Pratt's Peter Quill could lead the way around the Galaxy, as the Guardians try to track down one of the powerful Infinity Stones.
The only problem here is that the Avengers game announced by Sqaure Enix teased a brand new storyline, separate from the movies and comics. If they follow the same pattern, the movie lore could be absent from the game.

While Star Lord and company are the characters that fans will recognize, none of them were members of the original Guardians of The Galaxy.
The first team, which appeared in 1969, consisted of Vance Astro, Justice, Martinex, Charlie-27, and Yondu.
Many will remember Yondu as the Ravager leader from the films, and his original character was a hero in Marvel comics.
Using this character, the game could tie the story into the classic Guardians that comic readers remember. The only issue here is losing out on bankable characters like Groot.
However, there was a comic series called Guardians 3000, which saw the classic team travel through time. They eventually met up with the current Guardians team, and that could make for an interesting crossover game.

Square Enix created one of the most famous open-world RPG's in history - Kingdom Hearts. There's a great chance they could do it again with Guardians.
By sending someone like Peter Quill or Nova on a quest around the Galaxy, trying to collect the rest of their team members, the developer could capture the magic they created in the Kingdom Hearts franchise.
This also opens the game up to ship customization as it travels between planets, which would greatly benefit a property like Guardians.

Throughout the history of Marvel comics, Iron Man has fought alongside the Guardians Of The Galaxy.
If the Avengers game is a massive success - like many are predicting - they could use Iron Man as a bridge into the Guardians story. This would keep both properties existing in the same universe, and have fans wanting to purchase both games.
If they were to introduce Iron Man as a lead for the Guardians game, the studio could then use some of the older Guardians as playable characters. You wouldn't have to worry as much about having popular characters, when you've got Marvel's most bankable star on your roster.
