Lex Luthor
Obviously Lex Luthor will be at the head of the Legion of Doom, or Injustice League villain team, but fans want something better from this iconic mastermind than they saw in Batman v Superman. They want the smirking, maniacal Lex who can out-think anyone and is more methodical and strategic than even Batman, with less of the weird ticks seen in BvS.
Deathstroke
Slade Wilson usually starts as a villain before becoming a something more of an anti-hero, and there's already the setup for Joe Manganiello's Deathstroke to do the same. Slade Wilson would be a great enforcer for the villains team in Justice League 2 - and also the key figure who switches sides for noble reasons, and helps save the day.
Sinestro
One thing Lex would love to have, is his very own alien powerhouse on the team.Given Lex's brain-enhancing experience on the Kryptonian ship, it's safe to assume he has knowledge of the entire Green Lantern mythos, and perhaps how to reach out to one 'Fallen Lantern' in particular...
Assuming that Justice League 2 comes after the release of Green Lantern Corps, Sinestro and his yellow power ring could make a nice debut in The Injustice League gang. His fear-based power ring could also cause intense psychological harm to the Justice League team members.

Gorilla Grodd
Barry Allen dropped an Easter egg for this villain to make his DCEU debut, and Justice League 2 would be a great place to do it, if nowhere else. Grodd is a great addition to the Legion because he's always unpredictable, willing to betray Luthor in order to make his own grab for power or leadership of the team. With S.T.A.R. Labs officially part of the DCEU mythos now, explaining a super-intelligent psychic gorilla shouldn't be too hard.
Queen Clea
A more obscure choice of DC Villain, but solid in the fact that she's an exiled queen from a smaller kingdom within Atlantis, and is a classic Wonder Woman villain. That's one villain that could rival two key heroes of the Justice League.
Red Tornado
Characters like T.O. Morrow and Dr. Ivo have yet to make their presence felt in the DCEU, but one way to get that ball rolling (and eventually, get the League a new member, would be to introduce Morrow and his creation, the android Red Tornado, as part of Luther's team. Cyborg and Tornado could have an interesting rivalry, with Morrow also posing the threat of bending Cyborg's tech to his own nefarious control. That "man vs. machine" subplot could have a happy ending, with Cyborg sparking a noble sentience that turn Red Tornado into a Justice League hero.
Black Manta
This villain will make his debut in the Aquaman movie, but like Slade Wilson, his services can be bought for a price that Luther could easily pay. Manta is a master at ruthless strategy and subterfuge, and his unique status as an undersea pirate master make him (and his fleet) a threat that the Justice League may never see coming.
Black Adam
We've already heard that The Rock's Black Adam may appear in Suicide Squad 2, but the villain could get a much bigger (and pivotal) part to play as a member of Luther's team.
Adam is a powerful mystical deterrent to Superman's powers, and a formidable threat to Wonder Woman and Aquaman, as well. It could take the Justice League reaching out to hero like Shazam to finally stop him.

Bizarro
We've all seen how Lex Luthor loves to play mad scientist with Kryptonian DNA, and after the mistake of making an uncontrollable monster like Doomsday, he could perfect his experiment with a Bizarro Superman clone. The opportunity is already on the table: Superman was dead for awhile, leaving every opportunity for Luther to have procured Superman's DNA in some form or fashion. If nothing else, Lex would love the damage Bizarro could cause to Superman's reputation.
*Joker
Of course, The Joker is a headlining feature of various villain team-ups in the DC Universe, but we have to add the asterisk based on that same history. The problem with Joker is that he doesn't really play nice with others - for long, if at all. So while Joker could be approached by Lex, it's anyone's guess where and why he'd choose to wreak havoc on the Legion's behalf (if depicted right).
