You Don't Give Up
"We try to save as many people as we can. Sometimes that doesn't mean everybody but you don't give up."
Captain America admitting that he can't save everyone is a big step for him. He is a character who not only prides himself in helping and defending others, but also obligates himself to it. He looks at every single casualty with enormous weight.
For Captain America to argue against Tony Stark or the governing bodies implementing the Registration Act with a claim that he tries to save people but admitting he fails, he must truly despise the idea of being monitored by the new Accords.
It's a huge flip from the Captain America who started out as the S.H.I.E.L.D. "yes" man in The Avengers and anti-government Tony Stark from Iron Man.

Much like Captain America admitting that he can't save everyone, Tony Stark admitting that operating under government supervision is a huge flip for the character. This is the same guy who once mocked the U.S. government publicly and fought a hard fight to protect his technology from their hands.
The two characters have switched sides and have very strong beliefs in their new efforts. It's going to get messy.
If you pay close attention, the line preceding this one is, "That's why I'm here." This indicates that Tony Stark shows up unexpectedly in the same meeting where Thaddeus Ross is lecturing Rogers, Romanov, and Wilson. This takes us all the way back to the post-credits scene from The Incredible Hulk where Stark approaches Ross to recruit the Hulk and their relationship was formed.

Whether he's talking to Captain America or Bucky, Tony Stark is getting really serious about this fight. The way the new trailer frames it, Tony Stark's buddy James Rhodes is killed of by Captain America or a supporter of his and it prompts Tony to lose any belief that Cap has good in him.
Tony is not fighting against Captain America on his own but he's getting the whole world behind him as much as he can.

It should be no surprise that Iron Man has all the best lines here despite Civil War being a Captain America movie. He's always been the wittiest Avengers.
When Iron Man shoves Cap aside, much like he did in Avengers: Age of Ultron, he utters, "You've just started a war." In other words, "It's on." Not that he really had to say anything to make it clear but hearing him say those words to Steve Rogers is a comic fan's dream. These Marvel heroes are really about to go to war with one another.

Sure, the line is simple, but it's the first time we've ever heard from Tom Holland's Peter Parker/Spider-Man in the Marvel Cinematic Universe. The webslinger has finally come home to movies with his fellow Avengers and this is just the beginning.
The line should be appreciated, too, as you hear the young voice of Holland's. He's probably genuinely excited to be where he is, despite being thrown into a battle between super heroes. This is a teenager from New York City who once watched these guys save his very city. The youth and excitement can be heard in those two words.
