There's a surprising amount of comedy in this film, giving it a nice balance to the action and drama. Even when they are directly at odds, the two teams still recognize that hey, they're friends, and they're even still joking with each other. When Falcon and Captain America are taken into custody alongside Winter Soldier, their gear and uniforms are taken. Falcon's is humorously catalogued as "one bird costume," to which he takes great, and funny, offense.
"You Could At Least Recognize Me"

This was a big one. When Black Widow and Winter Soldier rumble (thanks to a fresh brainwashing - man, does every dude in Black Widow's life get brain washed and fight her? Hawkeye did, Hulk did, and now Winter Soldier! Weird.), she drops this little nugget: "You could at least recognize me!"
In the pages of Marvel Comics, Natasha and Bucky have a history together from their time in Russia's various spy programs. They even have a romantic past, as well as having pulled missions alongside one another. That's never really been hinted at in the Marvel Cinematic Universe before, and certainly not outright confirmed - yes, they know each other - like it was with this line. It certainly opens up some future storytelling options.
"I'm Shaking Your Hand For Too Long"

Another dose of humor, this time from Scott Lang, Ant-Man. When he meets Captain America, having had only one real super powered adventure, Scott is understandably a bit nervous. His meeting, well-chronicled via released preview clips, is a great, deep breath before the crazy beat down they're all about to both inflict and receive at the hands of former teammates at the airport.
"I Could Do This All Day"

Yes, this line appeared in a couple of trailers and TV spots, too, but that doesn't downplay its awesomeness. When Cap is being pounded by Iron Man, and clearly losing (especially after Bucky was... incapacitated), it looks like he's down and out. Tony warns him to stay down, but Cap, as always, gets back up, and replies, "I could do this all day," while wavering in his place.
It's an exact repeat of what young, scrawny Steve Rogers did and said when he was being beat down by bullies before he ever became Captain America. It perfectly illustrates how Cap has had his true strength on the inside all along.
"Let Them Try"

It's the final line before the end credits role. As T'Challa and Steve Rogers stand in front of a window overlooking the majesty of the former's kingdom of Wakanda in the mid-credits scene, they discuss the fate of Bucky "The Winter Soldier" Barnes. He's being put back into a cryo-sleep while T'Challa's formidable scientists search for a way to permanently cure him of his brainwashing at the hands of Hydra and the Soviets. When Cap warns Black Panther that if anyone, good or bad, finds out that Bucky's there, they'll come for him, the superhero king answers simply, "Let Them Try."
It's powerful, it's incredible setup and establishment of his character for his solo mission, and it is a line that seems to beg for more, the way Marvel fans are after hearing it.
